If the battery is at 100% and you are using it while it still being plugged in then yes. I am proof because I do this sometimes and my battery now can only hold 78% of its full charge (even though it says 100%, it goes down much faster than it should).
If the computer is dying and you still need to use it, plugging it in and still using it will not harm the battery.

You battery MUST be kept in at all times, for example, if your laptop was running on the mains and the battery was out, what would happen if there was a power failure...it would crash your laptop
